The analyses identify glyphosate targets that correlate with kidney injury and kidney cancer, revealing pathways with significant glyphosate-induced alterations, including the dysregulation of <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/23142151\/\">nitrogen metabolism<\/a> that leads to ammonia accumulation and <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/category\/diseasehealth-effects\/oxidative-stress\/\">oxidative stress<\/a>, both of which contribute to renal (<a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/index.php?s=kidney\">kidney<\/a>) damage and carcinogenesis (development of <a href=\"https:\/\/www.beyondpesticides.org\/resources\/pesticide-induced-diseases-database\/cancer\">cancer<\/a>).<\/p>\n<p>\u201cThis study provides a comprehensive investigation into the molecular mechanisms by which glyphosate may contribute to kidney injury and kidney cancer, employing an array of bioinformatics tools for target prediction, toxicity assessment, pathway enrichment analysis, molecular docking and molecular dynamics simulation,\u201d the researchers state. The results of the analyses and simulations highlight the molecular mechanisms underlying glyphosate&#8217;s nephrotoxic (damaging to kidneys) and carcinogenic (cancer-causing) effects.<\/p>\n<p><strong>Study Importance and Background<\/strong><\/p>\n<p>Glyphosate, known as a broad-spectrum systemic herbicide, has been used for agricultural and nonagricultural purposes for decades and is the most extensively used herbicide worldwide. This widespread use is largely due to its application to genetically engineered, glyphosate-tolerant crops. Both glyphosate and its main <a href=\"https:\/\/www.beyondpesticides.org\/resources\/pesticide-gateway\/what-is-a-pesticide#:~:text=the%20production%20process.-,Metabolites,-Metabolites%20are%20breakdown\">metabolite<\/a> (breakdown product), <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/category\/chemicals\/aminomethylphosphonic-acid-ampa\/\">aminomethylphosphonic acid<\/a> (AMPA), are detected in water, soil, and food, which then represent <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/29117584\/\">multiple pathways for exposure<\/a> to nontarget organisms, including humans.<\/p>\n<p>There is a <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/category\/chemicals\/glyphosate\/\">wide body of science<\/a> connecting glyphosate to threats to public health, wildlife, and the environment. Both research and <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/2024\/02\/bayer-monsanto-in-roundup-glyphosate-case-stung-with-largest-multi-billion-dollar-jury-award-turns-to-state-houses-to-stop-litigation\/\">litigation<\/a> provide evidence of the carcinogenic potential of glyphosate, as well as toxicity to kidneys. Studies show both acute and chronic effects of glyphosate exposure, \u201ctriggering oxidative stress, inflammation, and apoptosis [cell death] in renal cells, culminating in structural and functional kidney damage.\u201d (See scientific literature <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/34990202\/\">here<\/a>, <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/34830483\/\">here<\/a>, and <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/36030873\/\">here<\/a>.)<\/p>\n<p>Additional epidemiological research links glyphosate exposure with higher rates of <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/37858770\/\">renal diseases<\/a>, including chronic kidney disease (CKD) and renal cell carcinoma. \u201cBoth in vitro [in a test tube or petri dish] and in vivo [in a living organism] studies reveal glyphosate\u2019s capacity to disrupt mitochondrial function, modulate gene expression involved in detoxification, and generate reactive oxygen species (ROS), collectively contributing to renal cell injury,\u201d the researchers point out. (See <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/36614029\/\">here<\/a> and <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/31874349\/\">here<\/a>.) They continue, \u201cGiven these findings, there is an urgent need to unravel the molecular pathways by which glyphosate may drive kidney injury and cancer progression, especially under conditions of prolonged exposure.\u201d<\/p>\n<p><strong>Methodology<\/strong><\/p>\n<p>To better understand the mechanisms through which glyphosate can impact kidney function and cancer development, without the limitations seen in traditional toxicological approaches with complex substances like glyphosate, this study utilizes network toxicology as a \u201ctransformative framework for dissecting the complex biological interactions triggered by chemical exposures.\u201d<\/p>\n<p>As the authors explain, \u201cUnlike traditional toxicology, which often focuses on individual <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/37080529\/\">molecular pathways<\/a> or isolated targets, network toxicology employs systems biology to analyze the chemical\u2019s impact across interconnected molecular networks.\u201d This approach allows for analysis of glyphosate\u2019s cellular and systemic effects that result in deleterious impacts on health.<\/p>\n<p>Utilizing computational approaches allows for \u201cthe prediction and prioritization of potential interactions between small molecules like glyphosate and complex biological systems,\u201d the researchers state. They continue, \u201cSpecifically, methodologies such as <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/33450511\/\">molecular docking and simulation<\/a> analysis are employed to predict the binding affinity and favored orientation of glyphosate with proteins implicated in critical cellular processes\u2014particularly those related to renal function and carcinogenesis (e.g., key enzymes, receptors, or transporters identified from network analyses).\u201d For more information on the specific databases and computational tools within the study, see the <a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC12375789\/#:~:text=associated%20renal%20diseases.-,Methods,-Network%20toxicological%20analysis\">methodology section<\/a> for more details.<\/p>\n<p><strong>Study Results and Implications<\/strong><\/p>\n<p>Through network toxicological analyses and simulations, this study predicts glyphosate\u2019s ability to incite kidney toxicity and carcinogenicity, which adds to the current body of science with existing toxicity data that indicates that increasing doses of glyphosate heighten risks of adverse health effects. The results include:<\/p>\n<ul>\n<li>The prediction of 47 drug targets of glyphosate, 20 of which overlap with kidney injury and 31 with kidney cancer, represents how glyphosate can modulate specific pathways through key targets to cause diseases, shown through the creation of a \u201cdrug-target-pathway network.\u201d (See <a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC12375789\/#:~:text=Fig.%C2%A02A).-,Fig.%202,-.\">here<\/a>.)<\/li>\n<li>Additional predictions from databases indicate \u201cthat glyphosate\u2019s co-toxicity primarily involves kidney injury. Additionally, the databases predicted carcinogenicity, <a href=\"https:\/\/www.beyondpesticides.org\/resources\/pesticide-induced-diseases-database\/brain-and-nervous-system-disorders\">neurotoxicity<\/a>, <a href=\"https:\/\/www.beyondpesticides.org\/resources\/pesticide-induced-diseases-database\/asthma\">respiratory toxicity<\/a>, and ototoxicity [ear poisoning] for glyphosate.\u201d<\/li>\n<li>Analyses of protein-protein interaction networks involving glyphosate-induced effects show ten targets for glyphosate-induced kidney injury and ten targets for kidney cancer. These targets include <a href=\"https:\/\/www.sciencedirect.com\/topics\/medicine-and-dentistry\/matrix-metalloproteinase\">matrix metalloproteinases<\/a> (MMPs) that play crucial roles in processes such as tissue remodeling, wound healing, and embryonic development, as well as in diseases such as cancer and chronic inflammation. The authors note, \u201cWhile MMPs are known targets in <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/33589810\/\">cancer biology<\/a> their specific implication in glyphosate-induced renal toxicity represents a significant mechanistic advancement elucidated by our network-based approach.\u201d<\/li>\n<li>Gene Ontology analyses reveal prominent biological processes, including proteolysis (breakdown of proteins), extracellular matrix disassembly, negative regulation of apoptosis, and collagen catabolic process for glyphosate-induced kidney injury and cancer.<\/li>\n<li>Significant signaling pathways involved in glyphosate-induced kidney injury include bladder cancer, adherens junction (provides adhesion between cells), endocrine resistance, <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/30592984\/\">relaxin signaling pathway<\/a>, estrogen signaling pathway, fluid shear stress and <a href=\"https:\/\/www.mayoclinic.org\/diseases-conditions\/arteriosclerosis-atherosclerosis\/symptoms-causes\/syc-20350569#:~:text=Atherosclerosis%20is%20the%20buildup%20of,leading%20to%20a%20blood%20clot.\">atherosclerosis<\/a>, nitrogen metabolism, <a href=\"https:\/\/www.sciencedirect.com\/topics\/pharmacology-toxicology-and-pharmaceutical-science\/proteoglycan\">proteoglycans<\/a> in cancer, diabetic cardiomyopathy (heart muscle disease), and lipid and atherosclerosis. The pathways related to glyphosate-induced kidney cancer include nitrogen metabolism, metabolic pathways, bladder cancer, adherens junction, endocrine resistance, relaxin signaling pathway, estrogen signaling pathway, and <a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/shear-stress#:~:text=In%20the%20body%2C%20shear%20stress%20is%20mainly%20prominent%20in%20the%20blood%20vessels%20where%20the%20mechanical%20frictional%20forces%20created%20by%20the%20blood%20flow%20create%20a%20stress%20along%20the%20blood%20vessel%20wall.\">fluid shear stress<\/a> [frictional forces impacting blood flow] and atherosclerosis. As a note, nitrogen metabolism is the pathway with the <a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC6607905\/#:~:text=the%20ranked%20list.-,Pathway%20enrichment%20analysis,-.\">highest enrichment<\/a> [higher statistical significance] in both glyphosate-induced kidney injury and kidney cancer, which offers insight into less-explored mechanisms of glyphosate toxicity.<\/li>\n<\/ul>\n<p>\u201cIn summary, this study employed a robust computational strategy to propose that glyphosate may exert nephrotoxic and carcinogenic effects primarily through disrupting extracellular matrix integrity via interactions with MMPs and PLG [plasminogen, an enzyme involved in breaking down blood clots and other processes such as inflammation], and by dysregulating nitrogen metabolism,\u201d the researchers state. These results advance the mechanistic understanding of glyphosate toxicity, beyond general oxidative stress, which is crucial in understanding the health risks from both short-term and long-term exposure.<\/p>\n<p><strong>Previous Research<\/strong><\/p>\n<p><a href=\"https:\/\/www.beyondpesticides.org\/resources\/pesticide-gateway?pesticideid=37\">Studies upon studies<\/a> highlight the adverse effects associated with the widely used weed killer glyphosate, ranging from endocrine disruption and reproductive dysfunction to neurotoxicity and developmental impacts, among others. As shared in previous <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/2023\/08\/chronic-kidney-disease-of-unknown-origins-linked-to-indoor-pesticide-use-disproportionally-affecting-women\/\"><em>Daily News<\/em><\/a>, in 2013, the <a href=\"https:\/\/publicintegrity.org\/health\/new-urgency-targets-mysterious-kidney-disease-in-central-america\/\">Center for Public Integrity<\/a> highlighted that glyphosate bonds with toxic heavy metals in the environment, such as cadmium and arsenic, forming stable compounds. These compounds are present in food and water for consumption and do not break down until they reach the kidneys. Thus, <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/category\/farmworkers\/\">farmworkers<\/a> exposed to glyphosate are likely to have these toxic metals in their kidneys. Additionally, in 2019, researchers Sararath Guanatilake, MD, and Channa Jayasumana, PhD, were awarded the <a href=\"https:\/\/www.aaas.org\/sites\/default\/files\/2019-11\/AAAS%20Statement%20on%20the%202019%20Scientific%20Freedom%20and%20Responsibility%20Award.pdf\">Freedom and Responsibility Award<\/a> from the American Association for the Advancement of Science for <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/2012\/10\/kidney-disease-in-sri-lanka-linked-to-pesticides\/\">their work<\/a> in uncovering the link between glyphosate and chronic kidney disease. (See <em>Daily News<\/em> coverage <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/2019\/02\/researchers-awarded-for-uncovering-link-between-glyphosate-and-kidney-disease\/\">here<\/a>.)<\/p>\n<p>A previous<em> Daily News,<\/em> entitled \u201c<a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/2015\/09\/kidney-liver-damage-linked-to-chronic-low-dose-glyphosate-exposure\/\">Kidney, Liver Damage Linked to Chronic, Low-Dose Glyphosate Exposure<\/a>,\u201d highlights a research study published in <a href=\"https:\/\/ehjournal.biomedcentral.com\/articles\/10.1186\/s12940-015-0056-1\"><em>Environmental Health<\/em><\/a> that links chronic, ultra-low dose exposure to glyphosate in drinking water to adverse impacts on the health of the liver and kidneys. <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/2016\/07\/glyphosate-causes-changes-dna-resulting-chronic-disease\/\">Further research<\/a> finds glyphosate causes changes to DNA function, resulting in chronic diseases such as kidney and liver diseases, <a href=\"https:\/\/www.beyondpesticides.org\/resources\/pesticide-induced-diseases-database\/diabetes\">diabetes<\/a>, and <a href=\"https:\/\/beyondpesticides.org\/dailynewsblog\/category\/diseasehealth-effects\/parkinsons\/\">Parkinson\u2019s<\/a>. Multigenerational effects from glyphosate exposure are also documented, with research associating cancer development with DNA alterations through oxidative stress mechanisms. By forging ties with governments, nonprofits, and people who rely on these natural resources, we reduce the need for unnecessary pesticide use and protect public health and the environment. Beyond Pesticides provides hands-on services to the public and supports local action by: identifying and interpreting hazards; and, designing safe pest management programs. With the information provided by Beyond Pesticides, people may not only be able to make informed choices and adopt practices that protect themselves and their families from unnecessary exposure to pesticides, but they will be able to effect changes on community-wide pest management decisions and policies that govern pesticide use, such as pesticide uses in parks, schools, for community insect control and along roadsides.
