Study of Pesticide Mixtures and Intestinal Barrier Dysfunction Further Highlights Regulatory Deficiencies

(Beyond Pesticides, March 13, 2025) Study results published in Pesticide Biochemistry and Physiology “suggest that combined [pesticide] exposure may further amplify the toxicity and compromise the intestinal barrier.” The researchers studied the toxic effects of the insecticides abamectin and spirodiclofen, as well as the fungicide fluazinam, individually and in combination. In exposing mice and Caco-2 cells to these pesticides, the results show the disruption of intestinal functions and highlight the need to assess potential synergistic effects of pesticide mixtures as a part of the regulatory review process.  Effects on the intestinal barriers of mice represent a threat to “the first line of defense against the external environment,” the researchers say. The intestinal mucosal epithelial structure plays an important role in preventing harmful substances from entering the intestines and causing damage to cells.  [Caco-2 cells represent a human cell line, derived from a colon cancer patient’s tissue, and mimic the small intestine. Children’s Health Threatened as Rates of Pediatric Cancers are Linked to Agricultural Pesticide Mixtures

(Beyond Pesticides, March 4, 2025) A study in GeoHealth of pediatric cancers in Nebraska links exposure to agricultural mixtures with the occurrence of these diseases. The authors find statistically significant positive associations between pesticide usage rates and children with cancer, specifically brain and central nervous system (CNS) cancers and leukemia. “Our study is the first to estimate the effect of an agrichemical mixture on the pediatric cancer rate in Nebraska,” the study authors share. “One significant advantage of our study is that we identified the pesticide consistently applied over 22 years in Nebraska counties and then estimated the overall mixture effect of these pesticides on pediatric cancer.” The elevated effect of pesticide mixtures, a reality that is not evaluated in the U.S. Environmental Protection Agency’s (EPA) pesticide registration program, was reported in Oecologia (2008), documenting harm to amphibian populations even if the concentration of the individual chemicals is within limits considered acceptable. Lit Review Shows Depth of Research Linking Pesticide-Induced Effects on Immune System, Leading to Disease Outcomes

(Beyond Pesticides, January 15, 2025) There is robust scientific literature that unpacks the adverse human health effects of pesticide exposure, however immunological impacts do not receive adequate attention in regulatory review processes, according to an in-depth literature review. In a piece published in Frontiers in Immunology (2024) critiquing recent peer-reviewed scientific studies, as well as unpublished research produced by the Institute of Biology and Experimental Medicine in partnership with the National Scientific and Technical Research Council in Argentina, researchers assess immune system effects of pesticide exposure, which creates the conditions for deadly health conditions including various forms of cancer. The focus of this study, according to the authors, is “to critically review fundamental aspects of toxicological studies conducted on PPPs [Plant Protection Products] to provide a clearer understanding of the risks associated with exposure to these compounds on human health.” PPPs are pesticide products that contain more than one active ingredient, and can include synergistic ingredients that supercharge them alongside inert ingredients that pesticide companies are not legally required to disclose under the Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA), but are often manipulated biologically and chemically active. Sublethal Pesticide Exposure, Inadequately Regulated, Adversely Affects Insect Health, Study Finds

(Beyond Pesticides, December 5, 2024) A novel, comprehensive study published in Science reviews a library of 1,024 different chemicals (herbicides, insecticides, fungicides, and plant inhibitors) finding that, even at “sublethal” exposure levels, 57% of tested chemicals impact the behavioral and physiological health of house fly larvae. Mosquito and butterfly populations are also susceptible to long-term adverse effects at sublethal levels. Environmental and public health advocates continue to raise concerns about adverse effects resulting from the failure of the U.S. Environmental Protection Agency (EPA) to consider cumulative exposure (resulting in aggregate and synergistic effects) across different mechanisms of toxicity and different classes of pesticides, including at exposure levels below allowable levels set by the agency. Glyphosate Mixtures Show Lethal and Sublethal Effects to Embryos, Highlights Regulatory Deficiencies

(Beyond Pesticides, November 13, 2024) A study in Chemosphere, conducted by researchers from the Institute of Biochemistry and Molecular Biology in Germany, reveals the varied lethal and sublethal effects of different glyphosate mixtures through tests on the South African clawed frog, Xenopus laevis (X. laevis). After exposing embryos to four glyphosate formulations, mortality, morphological defects, altered heartbeat rate, and impaired heart-specific gene expression are observed. Glyphosate, an herbicide and popular weed killer in many Roundup® products, is one of the most commonly detected pesticides in waterbodies worldwide, threatening aquatic organisms and overall biodiversity. This study investigates the effects of Glyphosat TF, Durano TF, Helosate 450 TF, and Kyleo, four formulations containing glyphosate, as compared to the effects of pure glyphosate on embryonic development in amphibians. The formulations consist of varying concentrations of the active ingredient glyphosate, as well as other active and inert ingredients. The authors share that, “Glyphosat TF contains 34% glyphosate and 10–20% d-glucopyranose, while Durano contains 39–44% glyphosate and 1–5% N–N-dimethyl-C12-C14-(even numbered)-alkyl-1-amines. In Helosate most of the ingredients are listed – 50–70% glyphosate, 1–10% isopropylamine, 1–3% lauryl dimethyl betaine, 0.25–1% dodecyl dimethylamine. Study Finds Pesticide Product Labels Fail to Convey Toxic Effects to Consumers 

(Beyond Pesticides, August 28, 2024) The U.S. Environmental Protection Agency’s (EPA) pesticide labeling requirements fail to adequately communicate acute toxicity levels to the public, as evidenced in a recent study of consumers published last month in the journal Nature. After evaluating whether the current three “signal” words (CAUTION, WARNING, DANGER) on pesticide products adequately convey pesticide toxicity, the authors conclude that current labeling may result in “unintended adverse effects” because it does not “effectively communicate toxicity risks to consumers.” The signal words on pesticide labels, based on laboratory animal testing for determining lethal doses, are intended to protect users of the product from exposure that can kill through inhalation, skin absorption, and ingestion of the pesticide. However, the signal words do not warn about long-effects like cancer, neurological diseases, reproductive harm, as well as other adverse effects associated with pesticide exposure. Toxic Pesticides Increase Rates of Chronic Kidney Disease in Agricultural Communities

(Beyond Pesticides, August 22, 2024) A screening and analysis of 36 pesticides finds traces of these chemicals in patients with “unexplained chronic kidney disease” from agricultural communities in the Indian province of Uttar Pradesh. Researchers conducting this study, published in the Journal of Toxicology and Environmental Health Sciences, identify organophosphates, organochlorines, and pyrethroids as the main culprits, building on years of existing research pointing to adverse health impacts originating from bioaccumulation of pesticides after acute and chronic exposure. Chronic kidney disease (CKD) is a condition that afflicts one in ten adults internationally, based on a peer-reviewed analysis from The Lancet. The Centers for Disease Control and Prevention (CDC) estimate that one in seven U.S. adults (or roughly 35.5 million people) have CKD, with as many as nine in ten adults with the disease undiagnosed, based on 2023 data. Study Shows Importance of Testing Pesticide Mixtures to Determine Adverse Ecosystem Effects

(Beyond Pesticides, June 6, 2024) Researchers link pesticide exposure to behavioral effects on zebrafish (Danio rerio) larvae, signaling a larger issue for overall population and ecosystem effects. In a study published in Biomedicines, the authors conduct a multi-behavioral evaluation of the effects of three pesticides, both individually and as mixtures, on larvae. As the authors state, “Even at low concentrations, pesticides can negatively affect organisms, altering important behaviors that can have repercussions at the population level.” By analyzing effects on individual zebrafish with single compounds and mixtures, this study shows the dangers of pesticides in aquatic systems regarding synergy (a greater combined effect when substances mix) and the ripples created throughout entire ecosystems. Researchers from the Department of Morphology and Animal Physiology, as well as the Department of Physics, from the Rural Federal University of Pernambuco in Brazil collaborated with the Department of Biology at Indiana University in Indianapolis to perform the study. Bayer Loses Bid to Overturn Neonicotinoid Ban in Europe

(Beyond Pesticides, May 11, 2021) Last week, multinational agrichemical company Bayer Cropscience lost its bid to overturn a 2018 ban on bee-toxic neonicotinoids throughout the European Union. The ruling from the European Court of Justice rejected all grounds on which the company filed its appeal, noting, “It must be held that the arguments put forward by Bayer CropScience cannot, in any event, succeed.” In denying the appeal, the court ruled Bayer responsible for paying its own legal fees, as well as the fees of environmental organizations that intervened to defend the ban. Environmental groups are applauding the ruling, as it reinforces several important aspects of the EU’s pesticide policy that favor greater public health and environmental protections.
